It might be that the size of the instrument is the issue here. I don't know if you have enough ram, but try switching the 'DFD' (Direct From Disk) option to 'Sampler'. This should load the samples in memory.

Alternatively, it could be that you have the latency setting of your sound driver too low and audio isn't being processed fast enough to the sound card buffer for it to get out. In that case, set the latency higher.

It could also be that the output volume is too high and it's clipping. Try turning the volume down (either the master instrument volume, or the volume knob in the amplifier). There are LED meters there for you, but they're not necessarily 100% accurate so maybe you hit a peak so quickly that the meters don't register it.

(I'd turn the volume to 50%, turn your speakers up, and see if the clicks continue -- this would at least eliminate that possibility.)

Or the raw samples could possibly have clicks in them. Some cheap sample manufacturers aren't careful about this and don't chop their samples right, or don't create them right in the first place. You may have to use the Mapping Editor and Loop Editor to zero in on offending samples and creatively reprogram around them.

The previous suggestions (by Marc JX8P) are also good possibilities.
